Item High Bone Mass in Mice Lacking Cx37 Due to Defective Osteoclast Differentiation(American Society for Biochemistry and Molecular Biology, 2014-02) Pacheco-Costa, Rafael; Hassan, Iraj; Reginato, Rejane D.; Davis, Hannah M.; Bruzzaniti, Angela; Allen, Matthew R.; Plotkin, Lilian I.; Department of Anatomy & Cell Biology, School of MedicineConnexin (Cx) proteins are essential for cell differentiation, function and survival in all tissues with Cx43 being the most studied in bone. We now report that Cx37, another member of the connexin family of proteins, is expressed in osteoclasts, osteoblasts and osteocytes. Mice with global deletion of Cx37 (Cx37-/-) exhibit higher BMD, cancellous bone volume, and mechanical strength compared to wild type littermates. Osteoclast number and surface are significantly lower in bone of Cx37-/- mice. In contrast, osteoblast number and surface and bone formation rate in bones from Cx37-/- mice are unchanged. Moreover, markers of osteoblast activity ex vivo and in vivo are similar to those of Cx37+/+ littermates. sRANKL/M-CSF treatment of non-adherent Cx37-/- bone marrow cells rendered a 5-fold lower level of osteoclast differentiation compared to Cx37+/+ cell cultures. Further, Cx37-/- osteoclasts are smaller and have fewer nuclei per cell. Expression of RANK, TRAP, cathepsin K, calcitonin receptor, MMP9, NFATc1, DCSTAMP, ATP6v0d1 and CD44, markers of osteoclast number, fusion or activity, is lower in Cx37-/- osteoclasts compared to controls. In addition, non-adherent bone marrow cells from Cx37-/- mice exhibit higher levels of markers for osteoclast precursors, suggesting altered osteoclast differentiation. The reduction of osteoclast differentiation is associated with activation of Notch signaling. We conclude that Cx37 is required for osteoclast differentiation and fusion and its absence leads to arrested osteoclast maturation and high bone mass in mice. These findings demonstrate a previously unrecognized role of Cx37 in bone homeostasis that is not compensated for by Cx43 in vivo.Item Variability of in vivo reference point indentation in skeletally mature inbred rats.(Elsevier, 2014) Allen, Matthew R.; Newman, Christopher L.; Smith, Eric; Brown, Drew M.; Organ, Jason M.Reference point indentation (RPI) has emerged as a novel tool to measure material-level biomechanical properties in vivo. Human studies have been able to differentiate fracture versus non-fracture patients while a dog study has shown the technique can differentiate drug treatment effects. The goal of this study was to extend this technology to the in vivo measurement of rats, one of the most common animal models used to study bone, with assessment of intra- and inter-animal variability. Seventy-two skeletally mature male Sprague-Dawley rats were subjected to in vivo RPI on the region between the tibial diaphysis and proximal metaphysis. RPI data were assessed using a custom MATLAB program to determine several outcome parameters, including first cycle indentation distance (ID-1st), indentation distance increase (IDI), total indentation distance (TID), first cycle unloading slope (US-1st), and first cycle energy dissipation (ED-1st). Intra-animal variability ranged from 13-21% with US-1st and Tot Ed 1st-L being the least variable properties and IDI the most highly variable. Inter-animal variability ranged from 16% (US-1st) to 25% (ED-1st 31 and IDI). Based on these data, group size estimates would need to range from 9-18/group to achieve sufficient power for detecting a 25% difference in a two-group experiment. Repeat tests on the contralateral limb of a small cohort of animals (n=17) showed non-significant differences over 28 days ranging from -6% to -18%. These results provide important data on RPI variability (intra- and inter-animal) in rats that can be used to properly power future experiments using this technique.Item Implementation of a Journal Prototype for Pregnant and Parenting Adolescents(2014-10) Bute, Jennifer J.; Comer, Karen; Lauten, Kathryn M.; Sanematsu, Helen; Moore, Courtney M.; Lynch, Dustin; Chumbler, Neale R.Teenage pregnancy and childbearing remain pressing public health issues that have garnered attention from public health officials and social services agencies. This paper reports on the initial implementation and formative evaluation of a journaling program used as a means of communicating health information to pregnant and parenting adolescents (young women age 15-19) while also providing participants with a means of self-expression. The journaling prototype was implemented in a community-based agency in the Midwest by Family Support Specialists (FSSs) who made home visits on a monthly basis to assist pregnant and parenting adolescents (n = 52) with successful family planning and public health education. A mixed method approach of qualitative (analysis of journals, field notes, and responses of semi-structured interviews with FSSs) and quantitative (questionnaires from pregnant and parenting adolescent respondents) data with purposive sampling was employed to evaluate the implementation of the journaling intervention. Twenty of the 52 study participants were pregnant when the journaling intervention was implemented, while 32 were not pregnant, but recently had a child and were currently parenting. Two core themes emerged from analysis of the data after the implementation of the journals: (1) usefulness of the journal and responsiveness to participants' information needs and (2) functionality challenges. The results offer practical starting points to tailor the implementation of journaling in other contexts. Further, areas for improvement emerged regarding the distribution timeline for the journal and the content of the journal itself. As such, we discuss the lessons learned through this collaborative project and suggest opportunities for future phases of the journal intervention.Item Law Libraries as Publishers(2014-09) Keele, Benjamin J.Item Racial and Ethnic Diversity in Grounded Theory Research(Nova Southeastern University, School of Social and Systematic Studies, 2014) Burke Draucker, Claire; Al-Khattab, Halima; Hines, Dana D.; Mazurczyk, Jill; Russell, Anne C.; Stephenson, Pam Shockey; Draucker, ShannonNational initiatives in the United States call for health research that addresses racial/ethnic disparities. Although grounded theory (GT) research has the potential to contribute much to the understanding of the health experiences of people of color, the extent to which it has contributed to health disparities research is unclear. In this article we describe a project in which we reviewed 44 GT studies published in Qualitative Health Research within the last five years. Using a framework proposed by Green, Creswell, Shope, and Clark (2007), we categorized the studies at one of four levels based on the status and significance afforded racial/ethnic diversity. Our results indicate that racial/ethnic diversity played a primary role in five studies, a complementary role in one study, a peripheral role in five studies, and an absent role in 33 studies. We suggest that GT research could contribute more to health disparities research if techniques were developed to better analyze the influence of race/ethnicity on health-related phenomena.Item Enhancing Creativity in Teaching and Learning in Online, Face-to-Face and Hybrid Courses(2014-10-10) Hook, Sara Anne; Tennant, Felisa; Jones, Josette; Defazio, JosephThis engaging session will feature four faculty members from one school who have incorporated a number of pedagogical and technological approaches into their courses to encourage creativity in their students while continuing to nurture their own creativity as a way to stay motivated, innovative and engaged as teachers. It will include an interactive online activities for participants with an opportunity for self-reflection and illustrate some options for encouraging and assessing creativity in higher education. The session will review current research on creativity and distill the findings into practical applications for generating a learner-centered environment in any kind of classroom setting.Item Do Political Attitudes and Religiosity Share a Genetic Path?(Springer, 2014-10) Friesen, Amanda; Ksiazkiewicz, AleksanderSocial scientists have long recognized and sought to explain a connection between religious and political beliefs. Our research challenges the prevalent view that religion and politics constitute separate but related belief sets with a conceptual model that suggests the correlation between the two may be partially explained by an underlying psychological construct reflecting first principle beliefs on social organization. Moreover, we also push this challenge further by considering whether part of the relationship between political and religious beliefs is the result of shared genetic influences, which would suggest that a shared biological predisposition, or set of biological predispositions, underlies these attitudes. Using a classic twin design on a sample of American adults, we demonstrate that certain religious, political, and first principle beliefs can be explained by genetic and unique environmental components, and that the correlation between these three trait structures is primarily due to a common genetic path. As predicted, this relationship is found to hold for social ideology, but not for economic ideology. These findings provide evidence that the overlap between the religious and the political in the American context may in part be due to underlying principles regarding how to understand and organize society and that these principles may be adopted to satisfy biologically-influenced psychological needs.Item A Closed-Loop Model of the Respiratory System: Focus on Hypercapnia and Active Expiration(2014-10) Molkov, Yaroslav I; Shevtsova, Natalia A; Park, Choongseok; Ben-Tal, Alona; Smith, Jeffrey C; Rubin, Jonathan E; Rybak, Ilya ABreathing is a vital process providing the exchange of gases between the lungs and atmosphere. During quiet breathing, pumping air from the lungs is mostly performed by contraction of the diaphragm during inspiration, and muscle contraction during expiration does not play a significant role in ventilation. In contrast, during intense exercise or severe hypercapnia forced or active expiration occurs in which the abdominal “expiratory” muscles become actively involved in breathing. The mechanisms of this transition remain unknown. To study these mechanisms, we developed a computational model of the closed-loop respiratory system that describes the brainstem respiratory network controlling the pulmonary subsystem representing lung biomechanics and gas (O2 and CO2) exchange and transport. The lung subsystem provides two types of feedback to the neural subsystem: a mechanical one from pulmonary stretch receptors and a chemical one from central chemoreceptors. The neural component of the model simulates the respiratory network that includes several interacting respiratory neuron types within the Bötzinger and pre-Bötzinger complexes, as well as the retrotrapezoid nucleus/parafacial respiratory group (RTN/pFRG) representing the central chemoreception module targeted by chemical feedback. The RTN/pFRG compartment contains an independent neural generator that is activated at an increased CO2 level and controls the abdominal motor output. The lung volume is controlled by two pumps, a major one driven by the diaphragm and an additional one activated by abdominal muscles and involved in active expiration. The model represents the first attempt to model the transition from quiet breathing to breathing with active expiration. The model suggests that the closed-loop respiratory control system switches to active expiration via a quantal acceleration of expiratory activity, when increases in breathing rate and phrenic amplitude no longer provide sufficient ventilation. The model can be used for simulation of closed-loop control of breathing under different conditions including respiratory disorders.Item Flood disaster management policy: An analysis of the United States Community Ratings System(2014-10) Sadiq, Abdul-Akeem; Noonan, Douglas S.In 1990 the US Federal Emergency Management Agency created the Community Ratings System (CRS) to engage local governments to enhance community flood resilience. The CRS encourages community flood risk management activities by discounting flood insurance premiums commensurate with the level of flood management measures implemented. Using a national sample of communities, this study empirically identifies factors motivating both communities’ decision to participate and intensity of participation in the CRS. The results indicate that local capacity, flood risk factors, socio-economic characteristics, and political economy factors are significant predictors of CRS participation.
