Item Foundational Practices of Research Data Management(Pensoft, 2020-07-27) Briney, Kristin A.; Coates, Heather L.; Goben, AbigailThe importance of research data has grown as researchers across disciplines seek to ensure reproducibility, facilitate data reuse, and acknowledge data as a valuable scholarly commodity. Researchers are under increasing pressure to share their data for validation and reuse. Adopting good data management practices allows researchers to efficiently locate their data, understand it, and use it throughout all of the stages of a project and in the future. Additionally, good data management can streamline data analysis, visualization, and reporting, thus making publication less stressful and time-consuming. By implementing foundational practices of data management, researchers set themselves up for success by formalizing processes and reducing common errors in data handling, which can free up more time for research. This paper provides an introduction to best practices for managing all types of data.Item Inter-test reproducibility of the lung clearance index measured by multiple breath washout(ERS, 2017-10-01) Engberink, Esther Oude; Ratjen, Felix; Davis, Stephanie D.; Retsch-Bogart, George; Amin, Reshma; Stanojevic, Sanja; Pediatrics, School of MedicineThe lung clearance index (LCI) has strong intra-test repeatability; however, the inter-test reproducibility of the LCI is poorly defined. The aim of the present study was to define a physiologically meaningful change in LCI in preschool children, which discriminates changes associated with disease progression from biological variability. Repeated LCI measurements from a longitudinal cohort study of children with cystic fibrosis and age-matched controls were collected to define the inter-visit reproducibility of the LCI. Absolute change, the coefficient of variation, Bland–Altman limits of agreement, the coefficient of repeatability, intra-class correlation coefficient, and percentage changes were calculated. LCI measurements (n=505) from 71 healthy and 77 cystic fibrosis participants (aged 2.6–6 years) were analysed. LCI variability was proportional to its magnitude, such that reproducibility defined by absolute changes is biased. A physiologically relevant change for quarterly LCI measurements in health was defined as exceeding ±15%. In clinically stable cystic fibrosis participants, the threshold was higher (±25%); however, for measurements made 24 h apart, the threshold was similar to that observed in health (±17%). A percentage change in LCI greater than ±15% in preschool children can be considered physiologically relevant and greater than the biological variability of the test. Previous studies have demonstrated pQCT provides precise estimates that are good predictors of actual bone mechanical properties at popular distal imaging sites (tibia and radius). The predictive ability and precision of pQCT at more proximal sites remains unknown. The aim of the current study was to explore the predictive ability and short-term precision of pQCT estimates of mechanical properties of the midshaft humerus, a site gaining popularity for exploring the skeletal benefits of exercise. Predictive ability was determined ex vivo by assessing the ability of pQCT-derived estimates of torsional mechanical properties in cadaver humeri (density-weighted polar moment of inertia [IP] and polar Strength Strain Index [SSIP]) to predict actual torsional properties. Short-term precision was assessed in vivo by performing six repeat pQCT scans at the level of the midshaft humerus in 30 young, healthy individuals (degrees of freedom = 150), with repeat scans performed by the same and different testers and on the same and different days to explore the influences of different testers and time between repeat scans on precision errors. IP and SSIP both independently predicted at least 90% of the variance in ex vivo midshaft humerus mechanical properties in cadaveric bones. Overall values for relative precision error (root mean squared coefficients of variation) for in vivo measures of IP and SSIP at the midshaft humerus were less than 1.5% and were not influenced by pQCT assessments being performed by different testers or on different days. These data indicate that pQCT provides very good prediction of midshaft humerus mechanical properties with good short-term precision, with measures being robust against the influences of different testers and time between repeat scans.Item Quality control questions on Amazon's Mechanical Turk (MTurk): A randomized trial of impact on the USAUDIT, PHQ-9, and GAD-7(Springer, 2021-08-06) Agley, Jon; Xiao, Yunyu; Nolan, Rachael; Golzarri-Arroyo, Lilian; School of Social WorkCrowdsourced psychological and other biobehavioral research using platforms like Amazon's Mechanical Turk (MTurk) is increasingly common - but has proliferated more rapidly than studies to establish data quality best practices. Thus, this study investigated whether outcome scores for three common screening tools would be significantly different among MTurk workers who were subject to different sets of quality control checks. We conducted a single-stage, randomized controlled trial with equal allocation to each of four study arms: Arm 1 (Control Arm), Arm 2 (Bot/VPN Check), Arm 3 (Truthfulness/Attention Check), and Arm 4 (Stringent Arm - All Checks). Data collection was completed in Qualtrics, to which participants were referred from MTurk. Subjects (n = 1100) were recruited on November 20-21, 2020. Eligible workers were required to claim U.S. residency, have a successful task completion rate > 95%, have completed a minimum of 100 tasks, and have completed a maximum of 10,000 tasks. Participants completed the US-Alcohol Use Disorders Identification Test (USAUDIT), the Patient Health Questionnaire (PHQ-9), and a screener for Generalized Anxiety Disorder (GAD-7). We found that differing quality control approaches significantly, meaningfully, and directionally affected outcome scores on each of the screening tools. Most notably, workers in Arm 1 (Control) reported higher scores than those in Arms 3 and 4 for all tools, and a higher score than workers in Arm 2 for the PHQ-9. GABAergic dysfunction has been observed in the aging cerebellum. Studying cerebellar GABA changes is of considerable interest in understanding certain age-related motor disorders. However, little is known about the reproducibility of GABA MRS in an aged population. Therefore, this study aimed to explore the feasibility and reproducibility of GABA MRS in the aged cerebellum at 3.0 Tesla and to examine the effect of differing tissue composition on GABA measurements. MRI and 1H MRS exams were performed on 10 healthy elderly volunteers (mean age 75.2 years ± 6.5 years) using a 3.0 Tesla Siemens Tim Trio scanner. Among them, 5 subjects were scanned twice to assess short-term reproducibility. The MEGA-PRESS J-editing sequence was used for GABA detection in two volumes of interest (VOIs) in left and right cerebellar dentate.
