Item Advice Regarding Alcohol Use by Individuals With Nonalcoholic Fatty Liver Disease: Primum non nocere(Wiley, 2018) Dunn, Winston; Chalasani, Naga; Medicine, School of MedicineIndividuals with nonalcoholic fatty liver disease (NAFLD) often ask their health care providers about routinely consuming non‐heavy amounts of alcohol for pleasure and potential health benefits. According to the American Association for the Study of Liver Diseases (AASLD) Practice Guidance published in 2018, there are insufficient data to make a recommendation with regard to non‐heavy alcohol consumption by individuals with NAFLD.(1) Although several cross‐sectional studies have suggested that light alcohol consumption (on average, <1 drink per day) may have a beneficial effect on the presence and severity of NAFLD, a large meta‐regression analysis suggested that lower body mass index among light and moderate drinkers is a potential confounder and thus casts doubt on the rigor of those studies.Item Developmental Considerations for Assessment and Treatment of Impulsivity in Older Adults(Springer, 2020) Liu, Melissa; Argyriou, Eva; Cyders, Melissa A.; Psychology, School of ScienceImpulsivity is an important factor in many clinical disorders, especially alcohol and substance use disorders. Most of the research on impulsivity in this domain has focused on adolescence and young adulthood, as this developmental period is characterized by onset of and escalation in alcohol and substance use, likely driven in part by brain development patterns. Although many individuals eventually “mature out” of these behaviors in middle adulthood, a critical subset of people do not. The role of impulsivity in middle-to-older adulthood, when certain individuals transition from normative to disordered substance use, has not been carefully examined. The goal of this paper is to review the literature on measuring and modifying impulsivity from adolescence through older adulthood, with a special focus on middle-to-older adulthood. We propose that impulsivity research should include data on middle-to-older adulthood as an important time of transition to disordered use. We consider how impulsivity might have unique meaning at different stages of the adult lifespan and suggest modifications for assessing and treating impulsivity in older adults.Item AN EXAMINATION OF DEVELOPMENTAL AND SEX DIFFERENCES IN ETHANOL CONSUMPTION BY LOW ALCOHOL-CONSUMING RAT LINES(Office of the Vice Chancellor for Research, 2012-04-13) Largent, Tammie Rachell; Neal-Beliveau, Bethany S.; Bell, Richard L.In the United States, alcohol use and dependence is a major health issue affecting 4-5% of the population (Hasin et al., 2007). Research indicates ad-olescents ages 12-20 drink 11% of all alcohol consumed nationally, with more than 90% consumed in the form of binge drinking (Center for Disease Control and Prevention, 2010). Similar to the human condition, adolescent rodents generally consume more ethanol than their adult counterparts. Current rat animal model studies on alcoholism remain weighted toward examining Family History Positive (FHP), selectively bred, alcohol-preferring lines. Also, research has generally been focused on ethanol consumption be-havior of male rodents. However, female rodents tend to consume more al-cohol than male rodents (e.g., Adams et al., 1991). In addition, existing re-search on adolescent vs. adult alcohol abuse using “FHP” rats is not paral-leled by research with “Family History Negative” (FHN) rats, which might re-veal factors that prevent/protect an individual from excessive ethanol intake during this crucial stage of development. The purpose of this study was to evaluate ethanol consumption by male and female FHN, selectively bred, alcohol-nonpreferring rats during adoles-cence and adulthood. Studying adolescent vs. adult behavior may reveal de-velopmentally-specific, protective factors. Also, examining male versus fe-male behavior may reveal sex-by-development factors guarding against al-cohol abuse. Animals were placed in cages and assigned to experimental conditions defined by the following independent variables: line of rodent, rodent’s sex and age of ethanol exposure. The following dependent measures were exam-ined: changes in body weight as well as water and ethanol consumption. These measures were taken at least 5 days per week. We hypothesized that there would be elevated levels of ethanol con-sumption (g ethanol/kg body weight/day) in (a) adolescent vs. adult rats and (b) female vs. male rats. Future research might focus on gene and/or protein expression differences within certain nuclei of the brain’s reward neurocircuit between the FHP and FHN lines of rats. Currently, some data has been collected and statistically analyzed. Upon completion the study re-sults will be prepared for presentation and manuscript submission. Funded in part by the Indiana University-Purdue University Indianapolis, Undergraduate Re-search Opportunities Program (UROP)Item HiTOP-based Optimal Personalized Assignment to Abstinence from Alcohol: A Precision Medicine Approach(2024-08) Argyriou, Evangelia; Cyders, Melissa; Wu, Wei; Stewart, Jesse; Plawecki, MartinAbstinence from drinking has traditionally been the primary goal of alcohol use disorder (AUD) treatment; however, there is evidence that individuals respond differently when trying to be abstinent from alcohol. The main goal of my study was to use a novel precision medicine approach to optimize assignment to short-term abstinence from alcohol based on a variety of individual characteristics. The sample consisted of 97 moderate-to-heavy drinkers aged 21-35. A within-subjects design was employed where each participant completed two counter-balanced intravenous alcohol sessions (one following abstinence and one during usual drinking). For the primary aim of this study (N = 47), crossover generalized outcome weighted learning was used to estimate an optimal individualized assignment rule to short-term abstinence based on prescriptive factors, including HiTOP-relevant dimensions and other characteristics. For a secondary aim (N = 50), logistic regression was used to test whether the subgroups estimated by the optimal rule were associated with a set of genetic and behavioral factors related to AUD, and subjective perceptions to alcohol intoxication. Findings showed that an estimated rule with higher granularity – higher-specificity traits and demographics – led to lower alcohol consumption overall compared with one-size-fits-all rules (i.e., assigning everyone to abstinence or assigning no one to abstinence). The effect sizes of the difference were small-to-medium and fell short of statistical significance. Family history of AUD had a positive trend association with benefit from abstinence, with one standard deviation increase in family history of AUD being associated with twice as high odds of being assigned to abstinence. Due to the limited sample size, the results should be interpreted with caution. Study results provided preliminary evidence that an individualized assignment rule based on relatively simple and easily accessible individual characteristics can lead to lower alcohol consumption than that observed if everyone or no one was assigned to abstinence (i.e., one-size-fits-all approach). Genetic predispositions reflected in family history of AUD may be a potential mechanism linking the assessed prescriptive factors with abstinence response, which is worth further exploration.Item The interactive effects of alcohol cravings, cue reactivity, and urgency on college student problematic drinking(2013-12-10) Karyadi, Kenny Ananda; Cyders, Melissa A.; Mosher, Catherine E.; Stewart, Jesse C.; Grahame, Nicholas J.Prior research indicated a high prevalence of problematic drinking among college students, suggesting a need for more effective screening approaches and treatments. The tendency to act rashly in face of strong emotions (e.g. positive and negative urgency), alcohol cravings, and cue reactivity all have been associated with problematic alcohol use. However, no studies have examined the interactive effects of alcohol cravings, urgency, and cue reactivity. I hypothesized that alcohol cravings will be associated with greater problematic drinking among individuals with higher levels of (1) urgency, (2) cue reactivity, and (3) cue reactivity and urgency. Data were collected from a sample (final n = 240) of college students through a survey, which consisted of measures of urgency, problematic drinking, and alcohol cravings. On the survey, participants were also exposed to alcohol and non-alcohol pictures, after which they assessed their level of cue reactivity. Results were examined using multiple regression and simple slope analyses. Results provided partial support for our hypotheses. Alcohol cravings were associated with greater problematic drinking at lower levels of negative urgency (b = 7.36, p< 0.001). Furthermore, alcohol cravings were most strongly associated with problematic drinking at high levels of cue reactivity and low levels of negative urgency (b = 8.69, p<0.001), and at low levels of cue reactivity and high levels of positive urgency (b = 6.56, p<0.001). These findings emphasize the importance of considering urgency and cue reactivity in understanding how alcohol cravings create risk for problematic drinking.Item The relationship among depressive symptoms, urgency, and problematic alcohol and cannabis use in community adults(Elsevier, 2019-01) Um, Miji; Hershberger, Alexandra R.; Cyders, Melissa A.; Psychology, School of ScienceThis study examined alternative models for how negative and positive urgency influence the relationship between depressive symptoms and alcohol and cannabis use in a community sample. Methods Participants included adults (n = 675; age = 42.57, SD = 15.66; 65.0% female; 74.2% White) in the community sample from the Rockland Project by the Nathan Kline Institute. Path analyses were conducted. Results Negative urgency was a unique mediator in the relationship between depressive symptoms and problematic alcohol use (β =0.121, 95% CI = 0.060–0.182) and problematic cannabis use (β =0.120, 95% CI = 0.060–0.179). Negative and positive urgency significantly moderated the relationship between depressive symptoms and problematic cannabis use (negative urgency: β = 0.092, 95% CI = 0.040–0.145; positive urgency: β = 0.070, 95% CI = 0.022–0.119), such that the relationship was positive at high levels and negative at low levels of urgency. The patterns and levels of the relationships between depressive symptoms and cannabis use differed between negative and positive urgency. Neither urgency trait moderated the relationship between depressive symptoms and problematic alcohol use. Conclusions Despite being strongly related, negative and positive urgency have distinct roles in the relationship between depressive symptoms and problematic alcohol and cannabis use. Previous finding with younger samples that do not include both traits in the model at time generalize and at other times do not replicate, which warrants the continued examination of how these traits impart risk across the lifespan.Item The role of positive urgency in alcohol-related risk-taking: An experimental investigation(2021-12) Um, Miji; Cyders, Melissa A.; Boehm, Stephen; Stewart, Jesse C.; Zapolski, Tamika C.The relationship between positive urgency, a personality trait reflecting rash action during extreme positive emotional states, and risk-taking has previously been experimentally examined. However, how positive urgency is related to risk-taking while under the acute influence of alcohol has not been examined. The overarching goal of this dissertation was to generate behavioral evidence concerning how the interaction between positive urgency and alcohol consumption influences risk-taking via changes in emotional arousal. In this study, 59 community dwelling adults (mean age = 29.45 (SD = 10.96), 32.2% women, 78% White) completed mood induction procedures (positive or neutral) while consuming a beverage (alcohol or placebo) and then completed the Balloon Analogue Risk Task (BART) as a measure of risk-taking. The positive mood induction was effective in inducing high arousal positive emotions. Overall, study hypotheses were not supported; however, because of low power, effect sizes and patterns of relationship are reported. The relationship between positive urgency and risk-taking was positive and small in the positive mood condition but negative and small in the neutral mood condition. The alcohol group and the placebo group showed similar patterns of risk-taking that are positive and small. Finally, the relationship between positive urgency changes in emotional arousal was positive and small only in the positive/alcohol condition; however, there was no relationship between changes in emotional arousal and risk-taking. These findings suggest that, while changes in emotional arousal may result from a combination of positive urgency and alcohol consumption, it may not be a focal mechanism that explains the relationship between positive urgency and risk-taking. Further, positive urgency is a risk factor whether or not alcohol consumption is present. Although the small sample size limited the power to test the hypotheses, the effect size estimates obtained in this study provide preliminary data for a more properly powered future study. The pattern of findings suggests the viability of further developing the current positive mood induction to establish a lab-based paradigm for positive urgency and the use of a different experimental risk-taking task to examine positive emotion-based risk-taking.
